Pilote was born in Trois-Rivières, Quebec, on January 17, 1931. He began playing hockey at a young age and quickly developed a reputation as a skilled defenseman. In 1949, he joined the Quebec Citadelles of the Quebec Junior Hockey League. He played two seasons with the Citadelles, winning the league championship in 1951.

In 1951, Pilote was drafted by the Chicago Blackhawks. He made his NHL debut in the 1951-52 season and quickly became a regular on the Blackhawks' blue line. Pilote was a physical presence on the ice, but he also had great puck-handling skills and a booming shot.

Pilote helped the Blackhawks win the Stanley Cup in 1961. He was also a key member of the Blackhawks' "Dynasty Years" in the 1960s. The Blackhawks won the Stanley Cup three times in a row from 1961 to 1963. Pilote was named the Norris Trophy winner as the NHL's top defenseman in 1963 and 1964.

Pilote retired from the NHL in 1975. He played 17 seasons in the NHL, all with the Blackhawks. He finished his career with 160 goals and 441 assists in 1,068 games. He was inducted into the Hockey Hall of Fame in 1975.
